Can the NAV composite calibration on a Garmin GI 275 CDI be done airborne? The install manual says to calibrate the VOR with a NAV radio tester to generate a 0° FROM radial VOR signal. Would flying on this radial be close enough or should it be done when tuned to a VOT? I can receive a LOC on the ground at my airport so I should be able to calibrate the LOC.

As a pilot or A&P you can do a functional check. 91.171 explains your options, one such option is flying on an airway and identifying a ground reference.

However, if you find the calibration to be off, I believe a repair station sign off is necessary to resolve. A repair station would have a calibrated test set as required by the installation manual.

The install manual language is clear. "Use an appropriate NAV tester to generate a localizer signal with 0.155 DDM left or right, and tune the NAV radio to the test frequency.". Nothing about doing an in flight calibration, using a field VOT, nor any other acceptable or approved substitute.
